MI6 Agent Cole Barker, was undercover as a Fulcrum agent, when he stole a chip, and was in the process of recovering the reader from a Fulcrum contact when apprehended by Team Bartowski. After his true identity as an MI6 agent was revealed, he then assisted them with an effort to capture the device.

The plan was accidentally disrupted by Chuck, who took the chip in an attempt to prove himself equal to Cole, and Cole, Sarah, and Chuck were all captured by Fulcrum. Both Cole and Chuck then claim to the be the Intersect, and Chuck was able to avoid interrogation and torture by accidentally knocking himself out. A government team led by Casey then arrive to rescue them.

Cole was about to return to England  in Chuck Versus the Lethal Weapon, but he was intercepted en route by Fulcrum, who tortured him in an attempt to find out the identity of the Intersect,  and if he had any knowledge of Perseus. Cole managed to withstand the ordeal and to fight his way through a dozen Fulcrum agents to return to Castle.

He and Chuck were then restricted to Castle since they are actively being sought by Fulcrum, while Casey and Sarah infiltrate the party where Perseus is supposed to be. Though Chuck manages to identify Perseus, his handlers are captured by Fulcrum. Rather than call General Beckman for help, Cole suggests that he and Chuck go to rescue them. He coaches Chuck in the usage of a gun.

He took a bullet for Sarah, fired by Duncan, the enemy agent who tortured him. He later took a bullet for Chuck, too, when they tracked down Perseus.

At episode's end, he invites Sarah to go away with him, but she refuses. He is both a bit disbelieving as well as resigned to the obvious, "Who'd have thought it? Cole Barker loses the girl to Chuck Bartowski."

  • As a British spy, Barker hits most of the Bond parallels.
  • Barker is the first to reiterate the line, "Come with me if you want to live," a nod to the Terminator movies.
